As with most other RPG game, Wasteland 3 is chock full of quests to complete, people to meet, monsters to kill, and agonizing decisions to make.

- Welcome to Colorado, where the Rangers get ambushed and nearly wiped out.
- Rangers' Last Hope, where the Rangers receive a new base.
- Home Away from Home, where the Rangers settle in their new home and acquire their first companion.
- Cornered Rats, where the Rangers help save Colorado Springs' farm from the Dorseys.
- Unwelcome Guests, where the Rangers get their first taste of Broadmoor Heights.
- Full House, where the Rangers fill out the roster at Peterson Air Force Base.
- Colorado proper
- The Psychopath, The Zealot, The Traitor, where the Rangers meet the progeny of the Patriarch to various, often disastrous, results.
- Reagan Reborn, where the Rangers can lead to a rebirth of the Gipper (much to the wasteland's misery) and deal with Valor Buchanan.
- Raising a Little Hell, where the Rangers rescue hostages in Aspen and confront Victory Buchanan.
- Sand in the Gastank, where the Rangers upset the balance of power among the Plain gangs and face Liberty Buchanan and complete Gods and Clowns alongside Canary in the Coal Mine.
- One of Us, where the Rangers instead help Ironclad Cordite assume control of the gangs by completing King Cordite, Nobody's Fools, and The Gods Have Spoken.

The following quests were cut and left unused before release.
Air Force Academy Ruins[ | ]
- Energy Independence and Old World Power, where the Rangers can give Colorado Springs energy independence. This was later recycled into the Cult of the Holy Detonation DLC.
- Payback, related to Inspector Delgado.
Cheyenne Mountain[ | ]
The entire FADERs story arc was cut from the game before release.
- Old Soldiers Never Die, they just FADERs Away, where the Rangers confront the Faders.
- Homecoming, where the Rangers help Jane Doe.
Everest Hotel[ | ]
Despite being widely used during the Fig crowdfunding campaign, Everest wound up on the cutting room floor.
- Missing Persons, kicking off the story arc here.
- Fixer Upper, a minor quest to repair things around the Hotel.
- A New Business Plan, where the Rangers decide on a new plan for the hotel.
- Rebranding, a minor quest following up on the above.
Steeltown[ | ]
A majority of these quests were reworked into the Battle of Steeltown DLC.
- Diagnostic Pilgrimage, where the Rangers take Abigail's Buddy to the Dunes of Thought.
- Boiling Point, where the Rangers aid an insurrection against Abigail.
- Electric Dreams, where the Rangers decide the fate of the Computation Engine at Steeltown.
- Factory Fracas, where the Rangers resolve labor disputes.
- Kill Grille, where the Rangers recover a nuclear battery.
- Smoke and Meetings, where the Rangers meet Angela Deth at Steeltown.
